If going by LU, Piccadilly line to Barons Court, District line to Gloucester Road then Circle line to Great Portland Street would minimise the need to use stairs, but might take a while.

Alternatively, Heathrow Express to Paddington, then Hammersmith & City line. This would mean a bit of a walk at Paddington, and the need to cross over the footbridge, but would be quite quick, though more expensive than the first option.
