Whats the best way there on a sat night about 10 pm with the least walking or
going up stairs?

If going by LU, Piccadilly line to Barons Court, District line to Gloucester Road then Circle line to Great Portland Street would minimise the need to use stairs, but might take a while.

Alternatively, Heathrow Express to Paddington, then Hammersmith & City line. This would mean a bit of a walk at Paddington, and the need to cross over the footbridge, but would be quite quick, though more expensive than the first option.

Taxi - provided it's a properly licensed (black) London taxi. May not be
the cheapest (about £40-45 depending on traffic[1]), but almost
certainly the best ! :-)

[1] Shouldn't be too bad at the day/times you're asking about.

The snag is that Great Portland Street has no train service most weekends
at present.

Why is that?

Because of ongoing work to enlarge Kings Cross St Pancras station in
connection with the Channel Tunnel Rail Link, there are no trains on the
Circle and associated lines between Baker Street and Liverpool Street.
During the week before your journey, you need to check the situation at
http://tube.tfl.gov.uk/content/nexus/ (Scroll down to Longterm Information
/ Weekend Travel Changes)

The nearest alternative stations are Regents Park and Warren Street.
Alternative routes are Piccadilly to Piccadilly Circus, then Bakerloo to
Regents Park; or Piccadilly to Leicester Square, Northern to Warren Street.
However, both the interchanges involve steps.

Least hassle (but more expensive) would be Heathrow Express to Paddington
(£13 single), then a taxi to Great Portland Street which is not more than 2
miles.

Suggest travelling to Regents Park as Gt Portland St is closed weekends for
engineering work.
Heathrow to Piccadilly Circus then Bakerloo line to Regents Park.
Interchange involves 19 stairs up, subway then 21 stairs up according to the
booklet access to the underground.

Do the Picadilly and Victoria lines at Green Park have cross-platform
interchanges like many victoria platforms?
