If you take Heathrow Express through to Paddington, it's very quick but also very expensive. And Paddington to Southwark is across town. So you'd be just as well off getting the tube all the way and save some money. Word of warning - the tube can get very crowded as you get into Central London, particularly in peak hours (or rush hours as the locals call them), and it isn't easy to interchange at many stations when you have luggage.
