What is the closest tube or bus stop to the Eurostar gates @ Waterloo?

If I remember correctly, you can go to waterloo on the tube, then follow he
eurostar signs, this should pop you out right near the gates

The 159 stops under the bridge, too. And isn't there an escalator from
York Road?

the 211 and 507 stop in the Cab Road though and you can walk along to a
level entrance into the main station and then escalator or lift down
into Eurostar. Alternatively if you alight in York Road you can follow
the footpath round to the roundabout at the foot of the Cab Road and
then follow the taxi route into the Eurostar terminal and enter at
ground level there. This avoids steps although there is a bit of a walk.

From Waterloo Road you can simply take an escalator up to the main line
concourse and then escalator / lift down again to Eurostar.
